siRNA Transfection:
Scrambled-non-silencing siRNA: RNAi is a biological phenomenon where double-stranded RNA (dsRNA) specifically reduces the expression of its corresponding gene. Effective inhibition of specific gene expression is experimentally achieved by the transfection of small interfering RNA (siRNA), which binds to the RISC complex, leading to the degradation of target complementary mRNA molecules in the cell. However, when using scrambled-non-silencing siRNA, which serves as a control, the siRNA sequence is designed not to target any specific mRNA, thus not reducing gene expression. This type of siRNA is crucial for validating RNAi experiments by demonstrating that the observed effects are due to specific gene silencing and not off-target effects or cytotoxicity. The success of RNAi experiments depends on the efficient delivery of siRNA into cells via transfection of stable and functional siRNA molecules. Most DNA transfection reagents are unsuitable for siRNA, as they are sensitive to serum or can induce cytotoxicity, negatively impacting gene expression studies. Specialized transfection reagents have been developed and optimized for siRNA transfection, allowing efficient transfection even in the presence of serum, without compromising transfection efficiency or cell viability. Ready-to-use transfection protocols eliminate the need for extensive siRNA optimization experiments.
