VERO Cell Line:
The Vero lineage was isolated from kidney epithelial cells extracted from Cercopithecus aethiops (African green monkey). The lineage was developed by Yasumura and Kawakita in 1962. The original cell line was named “Vero” from “Verda Reno” (green kidney in esperanto). Vero cells are used for multiple purposes, including screening for the toxins, as a host cell line for growing virus, as a host cell line for eukaryotic parasites (Trypanosomatids.) The Vero cell lineage is aneuploid and continuous. It can be replicated through numerous cycles of division without becoming senescent. Aneuploidy means having an abnormal number of chromosomes. Unlike normal mammalian cells Vero cells are interferon-deficient. They don’t produce type 1 interferons when infected.
Figure 1. GAPD mRNA levels were quantified using real-time RT-PCR in the VERO cells transfected with siRNAs targeting GAPD or non-silencing siRNA. Forty-eight hours post-transfection, the cells were harvested and analyzed by real-time RT-PCR for GAPDH mRNA expression levels. Data were normalized against the 18S rRNA signal. Control samples were either mock-transfected or untreated. Values are normalized to untreated sample. Data are means ± SD (n=3).
Figure 2. Protein expression of GAPDH in VERO cells. DNA plasmid expressing GAPDH or siRNA targeting GAPDH were transfected into VERO cells following Altogen Biosystems transfection protocol. At 72 hours post-transfection the cells were analyzed by Western Blot for protein expression levels (normalized by total protein, 10 µg of total protein loaded per each well). Untreated cells used as a negative control.
