Transfection Efficiency:

MEF Cell Line:

The MEF cell line was derived from murine (Mus musculus) embryonic fibroblast cells. This cell line is useful for studying mammalian embryonic tissue, and could be used as a transfection host in research related to cell biology and molecular biology. These cells are commonly used as feeder cells for other cell types. Many MEF cell lines exist, each of which exhibit slightly different characteristics.

Primary cell cultures are used in biological and gene therapy studies and serve as important model systems that may more accurately represent the biology of normal cells. Many cultured cell lines, as well as the majority of primary cell cultures are able to be transfected with exogenous nucleic acids when appropriate transfection approaches are employed. Since the majority of transfection methods causes significant toxicity in primary cell cultures, optimizing this procedure, specifically the protocol and reagents to be utilized, is essential for developing an effective transfection strategies for a given cell type. ALTOGEN® Kits for primary cells and sensitive cell lines have been designed to have significantly lower cytotoxicity than other alternatives.

Figure 1. GAPD mRNA levels were quantified using real-time RT-PCR in the MEF cells transfected with siRNAs targeting GAPD or non-silencing siRNA. Forty-eight hours post-transfection, the cells were harvested and analyzed by real-time RT-PCR for GAPD mRNA expression levels. Data were normalized against the 18S rRNA signal. Control samples were either mock-transfected or untreated. Values are normalized to untreated sample. Data are means ± SD (n=5).
